Combined use of isopropylamine and trifluoroacetic acid in methanol-containing mobile phases for chiral supercritical fluid chromatography

An acidic or basic additive is chosen, depending on the nature of the compound. This work highlights the simultaneous use of the acidic additive trifluoroacetic acid (TFA) and the basic additive isopropylamine (IPA) in supercritical fluid chromatography for enantioseparations. To evaluate the combination of TFA and IPA, 59 chiral pharmaceutical compounds were analyzed on four polysaccharide-based chiral stationary phases (CSPs): Lux(®) Cellulose-1, Lux(®) Cellulose-2, Lux(®) Cellulose-4 and Lux(®) Amylose-2. The results show that an important increase in enantioselectivity of the chromatographic system can occur when combining trifluoroacetic acid and isopropylamine in the mobile phase (MP), compared to the individual use of these additives. However, the combination of isopropylamine and trifluoroacetic acid in a supercritical methanol-containing mobile phase can also lead to problems as a result of the formation of salt complexes between the two additives. Combining the additives trifluoroacetic acid and isopropylamine and taking the appropriate measures to avoid salt formation, i.e. reducing the additives' concentrations, can lead to simpler chiral SFC screening conditions that display even broader enantioselectivity.

Photodeposited silver nanoparticles for on-column surface-enhanced Raman spectrometry detection in capillary electrophoresis

[More] A new, simple photo-deposition method of silver nanoparticles induced by laser inside a fused-silica capillary is described and tested. Silver nanoparticles are immobilized using Ar-ion laser beam of a wavelength of 488 nm and power of 3.6 mW for 60 min. The photodeposited compact spot of a size of ∼10 μm is temporary and spatially stable and resistant to a hydrodynamic flow. The deposit has very good properties for surface-enhanced Raman scattering and serves well for detection in capillary electrophoresis. The advantage of this approach is that neither the silver nanoparticles nor the chemicals for their preparation are components of the background electrolyte during the electrophoretic separation. Thus, the substrate formation and separation of analytes are two independent processes and can be performed under their optimum conditions. The zone broadening due to the sorption of analytes on the immobilized nanoparticles can be significantly reduced by an addition of 20% solution of methanol. The efficiency of capillary electrophoresis and detection selectivity of surface-enhanced Raman scattering induced by He-Ne laser at 632.8 nm is demonstrated by the 3D electropherograms of rhodamines 123 and B as model samples. The limits of detection of about 49 and 150 fmol (1 and 2 μM) have been reached for rhodamine B and 123, respectively.

Determination of Low Levels of Methanol in Crude Oils by Multi-dimensional Gas Chromatography (MDGC) Using Novel Micro Channel Flow Technology

[More] The accurate and precise determination of methanol in crude oils at concentrations less than 10 ppm is of economic value to the petroleum industry. This report presents the optimization, results and long term performance of a flow switching device MDGC hardware, the Swafer™, for the rapid and precise analysis of methanol from approximately 0.4 ppm (w/w) to 1000 ppm. The use of low temperature injection and backflush technique decreases maintenance and increases sample throughput. The short term quantitative percent relative standard deviations at 1, 30 and 1000 ppm (w/w) methanol in crude oils are 5, 3 and 0.3, respectively. The MDGC procedure follows closely that described in ASTM D7059 for determination of crude oils; however, D7059 previously has not been evaluated at concentrations of less than 10 ppm. This work further extends the application of D7059 to concentrations of <10 ppm.

In this study, a simple and rapid thin layer chromatograph coupling with fluorometric (TLC-F) method has been used to determine the partition coefficient of target compounds in HSCCC solvent system. Two components, 6,7-dimethoxycoumarin and 5-hydroxymethyl-2-furfural were successfully separated from purple sweet potato extracts by successive sample injection for the first time, using n-hexane-ethyl acetate-methanol-water (1:2:1:1, v/v/v/v) as the solvent system. Additionally, statistical analysis showed that there was no significant difference in partition coefficient obtained by the TLC-F method and by HPLC, which demonstrated the usefulness of TLC-F method.

Density functional theory investigations on the structure and dissolution mechanisms for cellobiose and xylan in an ionic liquid: gas phase and cluster calculations

[More] Density functional theory (DFT) calculations have been carried out for cellobiose and xylan chosen as models for cellulose and hemicellulose, respectively, in gas phase, implicit and explicit solvent (water, methanol, and the ionic liquid, 1,3-dimethylimidazolium acetate) media using plane wave and atom centered basis set approaches in order to find out lowest energy conformers and configurations. Geometry, vibrational properties, and (1)H and (13)C NMR chemical shift values have been discussed under all three conditions. Calculations predict that inter- and intramolecular hydrogen bonding play an important role in the dissolution processes. In the gas phase and in implicit solvent, the anti-anti conformer of cellobiose and the anti-syn conformer of xylan are the most stable due to the formation of a large number of intramolecular hydrogen bonds. However, in the cluster calculations containing ion pairs of the ionic liquid (IL) surrounding the cellulosic units, the anti-syn conformer of cellobiose is more stable as intramolecular hydrogen bonds are substituted by intermolecular ones formed with the ions of the IL. The complexes of cellobiose (or of xylan) with the ions of the ionic liquid are stable with large negative binding energies ranging between -21 and -55 kcal mol(-1). The predicted (1)H NMR values of the lowest energy cellobiose conformers are in good agreement with the experimental value. Xylan binds stronger with the IL than cellobiose does by 20 kcal mol(-1). Furthermore, the two pentose rings in xylan are rotated by 60° to each other in contrast to their coplanarity in cellobiose, which can explain the higher solubility and the amorphous nature of hemicellulose in ionic liquids. The fewer number of hydroxyl groups in xylan (relative to cellobiose) does not affect the number of cations present in its first solvation shell while the number of anions is reduced.

Chitosan-functionalized carbon nanotubes as support for the high dispersion of PtRu nanoparticles and their electrocatalytic oxidation of methanol

[More] Carbon nanotubes (CNTs) were non-covalently functionalized with chitosan (Chit) and then employed as the support for PtRu nanoparticles. The functionalization was carried out at room temperature without the use of corrosive acids, thereby preserving the integrity and the electronic conductivity of the CNTs. Transmission electron microscopy reveals that PtRu nanoparticles were highly dispersed on the surface of Chit-functionalized CNTs (CNT-Chit) with small particle-size. Cyclic voltammetry studies indicated that the PtRu nanoparticle/CNT-Chit nanohybrids have a higher electrochemical surface area, electrocatalytic performance, and stability towards methanol oxidation compared to PtRu nanoparticles supported on the pristine CNTs.

Determination of accessible silanols groups on silica gel surfaces using microcalorimetric measurements

Based on the difference in immersion heats, a methodology for the determination of the number of silanols on the surface is presented. The calculated concentration of residual silanols on the silica gel surface agreed with data found in the literature. The proposed methodology, based on a calculation of possible hydrogen bond formation, was also tested on the series of bonded stationary phases with different coverage densities. A very good correlation between the calculated number of accessible residual silanols and the coverage density of bonded ligands was observed.

Short term effects of copper, sulfadiazine and difloxacin on the anaerobic digestion of pig manure at low organic loading rates

The influence of three frequently used antibiotics, copper dosed as CuSO(4), sulfadiazine (SDZ), and difloxacin (DIF), on the anaerobic digestion process of pig manure was studied in semi-continuous experiments. Biogas production recovered after every Cu dosage up to a sum of 12.94g Cukg(-1) organic dry matter (ODM), probably due to Cu precipitation following the formation of sulphide from sulphate. Complete inhibition was found at the very high Cu concentration of 19.40g Cukg(-1) ODM. Inhibitory effect of SDZ and DIF was observed at concentrations as high as 2.70gkg(-1) ODM and 0.54gkg(-1) ODM, respectively. It seems very unlikely that the antibiotics tested would inhibit the anaerobic process in a full-scale biogas plant.

Aqueous-methanolic extract of sweet flag (Acorus calamus) possesses cardiac depressant and endothelial-derived hyperpolarizing factor-mediated coronary vasodilator effects

[More] This investigation was aimed to probe the pharmacological base of medicinal use of Acorus calamus in ischemic heart diseases. Effect on heart parameters was studied in isolated rabbit heart while coronary vasodilator effect was studied in isolated bovine coronary arterial rings, suspended in tissue baths filled with Krebs solution, maintained at 37°C, aerated with carbogen and responses were measured on PowerLab data acquisition system. In Langendorrf's perfused rabbit heart, the crude extract of Acorus calamus (Ac.Cr) at 0.01-10 mg/mL partially suppressed force of ventricular contractions (FVC), heart rate (HR) and coronary flow (CF). The ethylacetate fraction completely suppressed FVC, partially suppressed HR and CF, while the nHexane fraction exhibited similar effect on FVC and HR but increased CF, similar to methacholine and arachidonic acid. In bovine coronary arterial preparations, Ac.Cr caused inhibition of U46619 (20 nM)-precontractions, which was blocked in presence of increasing concentration of K(+) (4.8-20 mM), tetraethylammonium (1 μM) and SKF525A (10 μM), similar to arachidonic acid and methacholine, indicating K(+) channels activation and possible involvement of endothelial-derived hyperpolarizing factor (EDHF). Activity-directed fractionation revealed that EDHF-mediated activity is concentrated in the nHexane fraction. When tested against high K(+), the ethylacetate fraction was found more potent than parent crude extract and nHexane fraction. These data indicate that Ac.Cr mediates coronary vasodilator effect primarily through EDHF, responsible for the increase in CF, while the cardiac depressant effects may be due to the presence of additional cardiac depressant constituent(s), thus provides possible mechanistic basis to its medicinal use in ischemic heart diseases.
