Factors affecting chelating extraction of Cr, Cu, and As from CCA-treated wood

[More] The disposal of chromated copper arsenate (CCA)-treated waste wood is becoming a serious problem in many countries due to potential leaching of hazardous elements from in-service use in the environment or disposal of solutions after remediation; therefore, it is necessary to develop proper remediation techniques. The effects of concentration, extraction period, temperature, and sequential extraction on the extraction of Cr, Cu, and As from CCA-treated wood using [S,S]-ethylenediaminedisuccinic acid (EDDS), ethylenediaminetetraacetic acid (EDTA), and nitrilotriacetic acid (NTA) were studied. Mobility of metal in the samples was evaluated by using a sequential extraction scheme that could give the information needed to explain different extraction efficiencies for different metals. Results of long-term leaching tests of CCA-treated wood before and after EDDS extraction were used to evaluate Cr, Cu, and As leachability. Kinetic experiments showed that 6 h was the optimum extraction time for all metals and CCA-treated wood. Experimental results showed that EDDS is a very effective chelating agent for the extraction of Cr, Cu, and As from CCA-treated wood. Increased temperature significantly enhanced the extraction efficiency of CCA metals, especially Cr and As. The much better extractability of Cu compared to Cr and As by chelating agents can be attributed to the presence of larger weakly bound fractions. The CCA-treated woods after EDDS extraction have met the EPA's TCLP regulatory limit and could be classified as a non-hazardous waste according to identification standard of hazardous wastes.

Arsenate accumulation and arsenate-induced glutathione export in astrocyte-rich primary cultures

To test whether arsenate affects the viability and the GSH metabolism of brain astrocytes, we have used primary astrocyte cultures as model system. Incubation of astrocytes for 2 h with arsenate in concentrations of up to 10 mM caused an almost linear increase in the cellular arsenic content, but did not acutely compromise cell viability. The presence of moderate concentrations of arsenate caused a time- and concentration-dependent loss of GSH from viable astrocytes which was accompanied by a matching increase in the extracellular GSH content. Half-maximal effects were observed for arsenate in a concentration of about 0.3 mM. The arsenate-induced stimulated GSH export from astrocytes was prevented by MK571, an inhibitor of the multidrug resistance protein 1. Exposure of astrocytes to arsenite increased the specific cellular arsenic content and stimulated GSH export to values that were similar to those observed for arsenate-treated cells, while dimethylarsinic acid was less efficiently accumulated by the cells and did not modulate cellular and extracellular GSH levels. The observed strong stimulation of GSH export from astrocytes by arsenate suggests that disturbances of the astrocytic GSH metabolism may contribute to the observed arsenic-induced neurotoxicity.

Mitochondrial hormesis links low-dose arsenite exposure to lifespan extension

[More] Arsenite is one of the most toxic chemical substances known and is assumed to exert detrimental effects on viability even at lowest concentrations. By contrast and unlike higher concentrations, we here find that exposure to low-dose arsenite promotes growth of cultured mammalian cells. In the nematode C. elegans, low-dose arsenite promotes resistance against thermal and chemical stressors, and extends lifespan of this metazoan, whereas higher concentrations reduce longevity. While arsenite causes a transient increase in reactive oxygen species (ROS) levels in C. elegans, co-exposure to ROS scavengers prevents the lifespan-extending capabilities of arsenite, indicating that transiently increased ROS levels act as transducers of arsenite effects on lifespan, a process known as mitohormesis. This requires two transcription factors, namely DAF-16 and SKN-1, which employ the metallothionein MTL-2 as well as the mitochondrial transporter TIN-9.1 to extend life span. Taken together, low-dose arsenite extends lifespan, providing evidence for non-linear dose-response characteristics of toxin-mediated stress resistance and longevity in a multicellular organism. © 2013 The Authors Aging Cell © 2013 Blackwell Publishing Ltd/Anatomical Society of Great Britain and Ireland.

Proteomic approach of adaptive response to arsenic stress in Exiguobacterium sp. S17, an extremophile strain isolated from a high-altitude Andean Lake stromatolite

[More] The North-Western part of Argentina is particularly rich in wetlands located in the Puna in an altitude between 3,600 and 4,600 m above sea level. Most of these high-altitude Andean lakes are inhospitable areas due to extreme habitat conditions such as high contents of toxic elements, particularly arsenic. Exiguobacterium sp. S17, isolated from stromatolites in Laguna Socompa, exhibited remarkable tolerance to high arsenic concentration, i.e., it tolerated arsenic concentration such as 10 mM of As(III) and 150 mM of As(V). A proteomics approach was conducted to reveal the mechanisms that provide the observed outstanding resistance of Exiguobacterium sp. S17 against arsenic. A comparative analysis of S17, exposed and unexposed to arsenic revealed 25 differentially expressed proteins. Identification of these proteins was performed by MALDI-TOF/MS revealing upregulation of proteins involved in energy metabolism, stress, transport, and in protein synthesis being expressed under arsenic stress. To our knowledge, this work represents the first proteomic study of arsenic tolerance in an Exiguobacterium strain.

XANES evidence of arsenate removal from water with magnetic ferrite

In this study, a magnetic ferrite, was synthesized and investigated for its As(V) removal efficiency. The adsorption of As(V) by magnetic ferrite exhibited an L-shaped nonlinear isotherm, suggesting limiting binding sites on the adsorbent surface. The As K-edge X-Ray Absorption Near-Edge Structure (XANES) revealed that the adsorbed As(V) on ferrite was not reduced to more toxic As(III) by Fe(2+) in the ferrite structure. The maximum As adsorption capacity of ferrite was 14 mg/g at pH 3 and decreased with increasing pH due to enhanced electrostatic repulsion between As(V) and the adsorbent surface. Desorption of As(V) using six different acid and salt solutions showed that the desorption rate decreased in an order of H3PO4 > Na3PO4 > H2SO4 > Na2SO4 > HCl > HNO3. These results suggest that magnetic ferrite without surface modification is an effective adsorbent for removing As(V) from water, which was confirmed by the effective removal of As(V) from contaminated groundwater using this material. The used material can then be recovered using a magnet because of its paramagnetism; the adsorbed As(V) on the material can be recovered using H3PO4 or Na3PO4 solutions.

Advantages of low pH and limited oxygenation in arsenite removal from water by zero-valent iron

[More] The removal of toxic arsenic species from contaminated waters by zero-valent iron (ZVI) has drawn considerable attention in recent years. In this approach, arsenic ions are mainly removed by adsorption to the iron corrosion products. Reduction to zero-valent arsenic on the ZVI surface is possible in the absence of competing oxidants and can reduce arsenic mobility and sludge formation. However, associated removal rates are relatively low. In the current study, simultaneous high reduction and removal rates of arsenite (H3AsO3), the more toxic and mobile environmentally occurring arsenic species, was demonstrated by reacting it with ZVI under limited aeration and relatively low pH. 90% of the removed arsenic was attached to the ZVI particles and 60% of which was in the elemental state. Under the same non-acidic conditions, only 40-60% of the removed arsenic was attached to the ZVI with no change in arsenic oxidation state. Under anaerobic conditions, reduction occurred but total arsenic removal rate was significantly lower and ZVI demand was higher. The effective arsenite removal under acidic oxygen-limited conditions was explained by formation of Fe(II)-solid intermediate on the ZVI surface that provided high surface area and reducing power.

Arsenite elicits anomalous sulfur starvation responses in barley

AsIII and S deficiency induced 5 - 20 fold increases in the three genes responsible for sulfate reduction. Both treatments also caused upregulation of a sulfate transporter, but only in the case of S deficiency was there an increase in sulfate influx. Longer term changes included reduction in transfer of S from roots to shoots, and an increase in root growth relative to shoot growth. Genes involved in complexation and compartmentation of As were upregulated by AsIII but not by S-deficiency. The rate at which As accumulated appeared to be controlled by the rate of thiol synthesis. Over a range of AsIII concentrations and growth periods, the ratio of thiols to As was always close to 3:1, which is consistent with the formation of a stable GS3:AsIII complex.The greater toxicity of As under S-limiting conditions is likely to be due to an intensification of S deficiency as a result of thiol synthesis rather than to a direct toxicity to metabolism. Since influx of AsIII was nearly 20-fold faster than the rate of synthesis of thiols, it is questionable whether this complexation strategy can be effective in preventing As toxicity unless As uptake becomes limited by diffusive resistances in the rhizosphere.

Arsenic speciation and spatial and interspecies differences of metal concentrations in mollusks and crustaceans from a South China estuary

[More] Arsenic speciation and concentrations were determined in mollusks and crustaceans in the intertidal zone from twelve locations in Zhanjiang estuary, South China. Metal concentrations (Ag, As, Cd, Cu, Hg, Ni, Pb, and Zn) were also concurrently determined in these species. Arsenic speciation analysis showed that the less-toxic arsenobetaine (AsB) constituted 80.6-98.8 % of all As compounds, and dimethylarsinic acid (DMA) constituted 0.47-3.44 %. Monomethylarsonic acid (MMA) and As(V) were only detected in the whelk Drupa fiscella and the crab Heteropilumnus ciliatus, respectively. Arsenite [As(III)] was not detected in any of the sampled specimens, but there were also unidentified other As species. A strong spatial variation of metals in the oyster Saccostrea cucullata was found in the estuary, confirming that oysters can be used as a good biomonitor of metal contamination in the studied area. The concentrations of eight metals in the studied mollusks and crustaceans clearly revealed that these invertebrates accumulated different metals to different degrees. Furthermore, As, Cd, Cu, Hg, and Pb contents in mollusks and crustacean samples were below the Food and Agricultural Organization (FAO) safe concentrations, thus there was no obvious health risk from the intake of the metals through marine mollusks and crustaceans consumption.

[More] ABSTRACT Nerve growth factor (NGF) is one of several structurally related proteins, named neurotrophins (NTs), that regulate neuronal survival, development, function, and plasticity. Moreover, NGF is an important activator of antioxidant mechanisms. These NGF functions are mediated by tropomyosin-related kinase receptor A (TrkA). Although NTs and their receptors have been shown to be expressed in visceral tissues, the extent to which NTs are involved in the physiology of visceral tissues is less clear. NGF is the most expressed NT in adult mouse livers. Although NGF is an important modulator of antioxidant mechanisms in neural tissues, few studies describe the relationship between oxidative stress and NGF expression in the liver. In the present study, we demonstrate that ngfb mRNA is positively modulated in mouse livers after oxidative injury via intraperitoneal injection of 14 mg/kg sodium arsenite, 6 mmol/kg L-buthionine-S-R-sulfoximine (BSO) or 300 mg/kg acetaminophen (APAP). In addition to the up regulation of ngfb we observed the phosphorylation of the NGF high-affinity receptor TrkA in the liver as well as downstream phosphorylation of Akt, NF-kB nuclear migration and iκbα and tx-1 mRNA up regulation. These effects were abolished when a neutralizing anti-NGF antibody was used. Furthermore, this anti-NGF antibody alone induced oxidative stress in the liver by decreasing reduced glutathione, increasing oxidized glutathione and down regulating tx-1 mRNA. Thus, NGF plays a critical role in liver protection against oxidative stress and xenobiotic injury as well as maintaining a reduced thiol state.

Arsenic Toxicity on Ludwigia octovalvis in Spiked Sand

The plants tolerated As(V) levels of 39 mg kg(-1) for as long as 35 days of exposure. After 91 days, the maximum concentration of As uptake in the plant occurred at As(V) concentration of 65 mg kg(-1) while As concentration in the stems, roots and leaves were 6139.9 ± 829.5, 1284.5 ± 242.9 and 1126.1 ± 117.2 mg kg(-1), respectively. In conclusion, As(V) could cause toxic effects in L. octovalvis and the plants could uptake and accumulate As in plant tissues.
