Prenatal exposure to perfluoroalkyl and polyfluoroalkyl substances and the risk of hypertensive disorders of pregnancy

However, epidemiological evidence on the association between PFAS and preeclampsia is inconsistent. We aimed to investigate the association between prenatal PFAS exposure and hypertensive disorders of pregnancy (HDP) in humans.

METHODS: PFAS were measured by liquid chromatography system coupled with tandem mass spectrometry in 687 umbilical cord plasma samples collected between 2011 and 2012 in Shanghai, China. Information on HDP including gestational hypertension and preeclampsia was abstracted from medical records. Multiple logistic regression was used to examine the association of each PFAS with gestational hypertension, preeclampsia, and overall HDP in separate models. Elastic net regression with logit link was used to identify independent associations between exposures and outcomes. Logistic regression was used to obtain the unpenalized estimates of the selected PFAS components for the associations with outcomes, adjusting for age, education level, pre-pregnancy BMI, parity, and mutual adjustment of selected PFAS.

RESULTS: The risk of gestational hypertension and preeclampsia was 3.3% and 2.8% in our subjects, respectively. Perfluorobutane sulfonate (PFBS), perfluorohexane sulfonate (PFHxS), perfluoroundecanoic acid (PFUA) were associated with preeclampsia based on elastic net penalty regression. In the fully adjusted statistical model, women with a higher level of standardized ln-transformed PFBS had an increased odds of preeclampsia [adjusted odds ratio (AOR): 1.81, 95% confidence interval (CI): 1.03-3.17], and overall HDP (AOR: 1.64, 95% CI: 1.09-2.47).

CONCLUSIONS: Prenatal exposure to PFBS was positively associated with the risk of preeclampsia and overall HDP.

Calibration and application of passive sampling for per- and polyfluoroalkyl substances in a drinking water treatment plant

[More] The aim of this study was to calibrate and apply polar organic chemical integrative samplers (POCIS) to examine 26 per- and polyfluoroalkyl substances (PFASs) in a drinking water treatment plant (DWTP). As a first step, the sampling rates (Rs) of 14 PFASs were determined in a laboratory calibration study for POCIS-WAX (weak-anion exchange) and POCIS-HLB (hydrophilic-lipophilic balance) (each with a surface area per mass of sorbent ratio of 227 cm2 g-1). While most PFASs were still in the linear uptake phase during the 28-day calibration study, Rs ranged from 0.003 to 0.10 L d-1 for POCIS-WAX and 0.00052 to 0.13 for POCIS-HLB. It is important to note that POCIS-WAX had higher Rs for short-chain perfluoroalkyl carboxylates (PFCAs) with a perfluorocarbon chain length of C3-C6 and perfluorobutane sulfonate (PFBS) compared with POCIS-HLB. Furthermore, Rs was significantly positively correlated with the sorbent-water partition coefficient (Kpw) for POCIS-WAX and POCIS-HLB (p < 0.0001). Use of POCIS-WAX and POCIS-HLB in the DWTP showed good agreement with composite water sampling. No removal of PFASs was observed in the full-scale DWTP. Overall, this is the first study of PFAS monitoring in a DWTP using two types of POCIS. The results demonstrate high suitability for future applications.

Prenatal exposure to perfluorobutanesulfonic acid and childhood adiposity: A prospective birth cohort study in Shanghai, China

[More] BACKGROUND: Several per- and polyfluoroalkyl substances (PFAS) have been phased out due to their adverse effects, and replaced by the short-chain perfluorobutanesulfonic acid (PFBS). However, the long-term impacts of PFBS on human health are unknown.

OBJECTIVE: We aimed to investigate the association between prenatal exposure to PFAS, especially PFBS and childhood adiposity at 5 years of age.

METHODS: We conducted a prospective birth cohort study involving 1,140 pregnant women from 2012 to 2017 in Shanghai. Fetal umbilical cord blood was collected at birth. A total of 404 children (196 girls) completed the adiposity measurements using a bioelectrical impedance analysis method and cord plasma PFAS measurements using LC-MS/MS. Multivariable linear models after adjustment for potential confounders were used to evaluate the associations between PFAS and childhood adiposity.

RESULTS: The median concentration of PFAS in the cord plasma ranged from 0.05 (PFBS) to 6.74 ng/mL (PFOA). Results of multivariable linear regression found that in girls, PFBS had a significant positive association with waist circumference and waist to height ratio (P-values < 0.05). Girls in the highest tertile of PFBS concentrations had more fat mass, as well as higher body fat percentage, waist circumference, and waist to height ratio compared to those in the lowest tertile. However, girls in the second tertile of PFDoA had lower body fat percentage, waist circumference and fat mass.

CONCLUSIONS: Adiposity at 5 years of age shows a positive association with prenatal exposure to PFBS in girls. These findings need to be further verified in larger prospective studies.

Highly Defective UiO-66 Materials for the Adsorptive Removal of Perfluorooctanesulfonate

While its use in most countries has been restricted to certain industrial applications due to environmental and health concerns, chrome plating and semiconductor manufacturing facilities are industrial point sources of PFOS-containing wastewater. Current remediation technologies are ineffective at treating these highly concentrated industrial effluents. In this work, UiO-66 metal organic frameworks (MOFs) of several defect concentrations were studied as sorbents for the removal of PFOS from concentrated aqueous solutions. PFOS sorption isotherms indicated that defective UiO-66, prepared with HCl as a modulator, had a maximum Langmuir sorption capacity of 1.24 mmol/g, which was similar to 2X greater than powdered activated carbon (PAC), but similar to 2X less than that of a commercial ion-exchange resin. Defective UiO-66 adsorbed PFOS 2 orders of magnitude faster than the ion-exchange resin. Large pore defects (similar to 16 and similar to 20 angstrom) within the framework were critical to the increased adsorption capacity due to higher internal surface area and an increased number of coordinatively unsaturated Zr sites to bind the PFOS head groups. Of the common co-contaminants in chrome plating wastewaters, chloride ions have a negligible effect on PFOS sorption, while sulfate and hexavalent chromium anions compete for cationically charged adsorption sites. These materials were also effective adsorbents for the shorter-chain homologue, perfluorobutanesulfonate (PFBS). The enhanced PFOS and PFBS adsorptive properties of UiO-66 highlight the advantage of structurally defective MOFs as a water treatment approach toward environmental sustainability.

Comprehensive analysis of photoinitiators and primary aromatic amines in food contact materials using liquid chromatography High-Resolution Mass Spectrometry

[More] A comprehensive strategy for the analysis of UV-ink photoinitiators and primary aromatic amines (PAAS) in food-packaging materials such as, juice tetrabricks, pouches and bags has been developed using liquid chromatography coupled to Orbitrap High-Resolution Mass Spectrometry (LC-Orbitrap-HRMS). The methodology includes both quantitative target analysis and post-run target screening analysis. The quantitative method was validated after a previous optimisation of the single-stage Orbitrap fragmentation through the Higher-Energy Collisional Dissociation (HCD) Cell. Overall, the quantitative method presented recoveries ranging from 78% to 119%, with a precision (RSD) lower than 20%, for the 18 substances in the scope of the target method. Limit of quantification (LOQ) for UV-inks photoinitiators ranged from 0.5 µg/kg-1 for Isopropyl Thioxanthone (ITX) and 2-Ethylhexyl 4-(dimethylamino) benzoate (EHDAB) to 5 µg/kg-1 for the rest of photoinitiators. LOQ for PAAs were 2 µg/kg-1 except for aniline (ANL) and 3,3' dimethylbenzidine (3,3'-DMB) which was 2.5 µg/kg-1 in the two studied simulants (acetic acid 3% and ethanol 50%). For post-run target screening a customized theoretical database, that included Bisphenols, Polyfluorinated compounds (PFCs), Phosphorus Flame Retardants (PFRs) and other substances was built. For identification purposes, a mass accuracy lower than 5 ppm, and some diagnostic ions including isotopes and/or fragments were used. The strategy was applied to 18 samples collected in the Valencian region (Spain). No compounds were detected when the standardised migration test was applied. However, in the destructive test, benzophenone and EHDAB were determined from tetrabrick and pouch materials. In the post-run target analysis two PFCs (Perfluorooctanoic acid and Perfluoro-1-butanesulfonate) and four PFRs (2-ethylhexyl diphenyl phosphate, tris(2-choloroisopropyl) phosphate, triphenyl phosphate and 2-ethylhexyl diphenyl phosphate) were identified.

Efficiency of maternal-fetal transfer of perfluoroalkyl and polyfluoroalkyl substances

However, the efficiency and influencing factors of PFASs maternal-fetal transfer remain unclear. We measured the levels of six perfluoroalkylcarboxylates, three perfluoroalkylsulfonates, and one sulfonamide in 369 pairs of maternal and umbilical cord serum and examined the transplacental transfer efficiency (TTE) of PFASs by the functional group and carbon chain length in a prospective birth cohort in Shandong, China. All ten PFASs were detected in both maternal and umbilical cord serum in nearly all samples. Maternal and cord levels were closely correlated (the correlation coefficient [r] ranging from 0.485 to 0.908) in most PFASs except perfluorobutane sulfonic acid (PFBS) (r = 0.159). TTE was significantly affected by the functional group and carbon chain length. Compared to perfluoroalkylcarboxylates, perfluoroalkylsulfonates had a lower ratio of maternal to fetal transfer. A U-shaped relationship between carbon chain length and TTE was observed for perfluoroalkylcarboxylates while a monotonic descending trend was identified between TTE and the increasing carbon chain length for perfluoroalkylsulfonates. PFASs can readily pass through the placenta. The functional group and carbon chain length are important determinants for the TTE of PFASs.

Length-specific occurrence and profile of perfluoroalkyl acids (PFAAs) in animal protein feeds

However, the occurrence of PFAAs has been rarely evaluated in various APFs, especially for emerging short-chain PFAAs. In the present study, we collected the most prevalent APFs (blood meal, meat meal, feather meal, soybean meal and DDGS) and found that Σ16PFAAs ranged from undetectable to 37.1 ng/g dry weight (dw) (average: 7.23 ng/g dw). Blood meal contained the highest PFAA levels, and results revealed that some terrestrial-derived APFs might be present at levels on par with the marine-derived feed. Animal-derived APFs (mean 10.9 ng/g dw) possessed higher ∑PFAAs than plant-derived APFs (0.75 ng/g dw). Length-specific PFAA profiles were specified between terrestrial-origin and marine-origin feeds in addition to animal-derived and plant-derived feeds. Short-chain PFAAs (PFBA, PFBS and PFHxS) were primarily found in blood meal, meat meal, soybean meal and DDGS, while the long-chain counterparts dominated in feather meal. It is of great concern that feed exposure to emerging contaminants has not drawn enough attention.

The occurrence of perfluoroalkyl acids in an important feed material (fishmeal) and its potential risk through the farm-to-fork pathway to humans

[More] Few studies have attempted to elucidate the occurrence of perfluoroalkyl acids (PFAAs) in animal feeds and how they play a role in human ingestion. Fishmeal was the most important animal-derived feed in global husbandry and may have been subjected to PFAA contamination considering the PFAAs' ubiquitous distribution in aquatic ecosystems. We collected ninety-two commercial fishmeal from the most important fishmeal-producing countries and found that Σ16PFAAs ranged from 0.65 to 85.5 ng/g (average: 18.2 ng/g, 12% moisture). PFOS still predominated, with unexpected high detection of PFUnDA. The wide occurrence of short-chain PFAAs (e.g., PFBA, PFBS) in fishmeal were found for the first time. From a geographical view, PFAA levels in fishmeal that originated from the Northern Hemisphere were significantly higher than those from the Southern Hemisphere (p < 0.01). Higher levels of under-studied long-chain PFAAs (PFNA, PFDA, PFUnDA, PFDoDA and PFTrDA) weighted more in industrialized areas than less industrialized areas, whereas the legacy PFAAs (PFOS, PFOA and PFHxS) were comparable among all regions. The estimated daily intake was calculated from animal feed to humans (via cultured fish) from 20 Chinese provinces by the Monte Carlo Simulation. A proportion of 29.8% of residents from the Fujian Province exceeded the EFSA's suggestion for PFOS ingestion.

Characterization of perfluoroalkyl substances in sediment cores from High and Low Arctic lakes in Canada

In this study, historical inventories of twenty-three PFASs (i.e. C4-C14, C16 perfluoroalkane carboxylic acids (PFCAs); C4, C6-C8, C10 perfluoroalkane sulfonic acids (PFSAs); perfluoro-4-ethyl-cyclohexane sulfonic acid (PFECHS); dodecafluoro-3H-4,8-dioxanonanoic acid (ADONA); 8-chloro-perfluoro-1-octane sulfonic acid (8-Cl-PFOS); chlorinated polyfluorinated ether sulfonic acids (Cl-PFESAs) including 9-chlorohexadecafluoro-3-oxanonane-1-sulfonic acid (6:2 Cl-PFESA) and 11-chloroeicosafluoro-3-oxaundecane-1-sulfonic acid (8:2 Cl-PFESA); as well as perfluorooctane sulfonamide (FOSA)) are determined in two intact sediment cores collected from Lake Hazen, located in northern Ellesmere Island at 82° N in 2012 and Lake B35, located in central Nunavut at 64° N in 2009. In Lake Hazen, fluxes of perfluorooctanoic acid (PFOA), perfluorodecanoic acid (PFDA), perfluorobutane sulfonic acid (PFBS), and perfluorooctane sulfonic acid (PFOS) increased during 1963-2011. In Lake B35, fluxes of perfluoroheptanoic acid (PFHpA), PFOA, perfluorononanoic acid (PFNA), and perfluoroundecanoic acid (PFUnDA) increased during 1952-2009. The temporal trends for PFASs in Lake Hazen and Lake B35 sediments are consistent with the continuous annual delivery of PFASs to the Arctic of Canada. Temporal trends in sediment cores appear to follow historical market changes in PFAS manufacturing inventory. The doubling time of PFAS fluxes are faster in Lake Hazen sediments than Lake B35 sediments. In Lake Hazen, this may be attributed to the enhanced delivery of sediment and historically-archived PFASs promoted by climate-induced glacier melting in the Lake Hazen watershed post-2005. Exponentially increasing PFAS temporal trends in High and Low Arctic lakes in Canada stress the importance of developing effective global regulatory policies for PFAS manufacturing and highlights the potential for climate change-induced contaminant release from melting glaciers in the Arctic.

Effects of Perfluoralkyl Substances on a Multigenerational Scale: A Case Study with Chironomus riparius (Diptera, Chironomidae)

[More] A multigenerational test with Chironomus riparius was performed to assess long-term effects on life-traits of exposure to selected perfluoroalkyl compounds: perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), and perfluorobutane sulfonate (PFBS). These persistent contaminants are widespread in aquatic ecosystems at low concentrations, possibly exerting long-term toxicity. Larvae of C. riparius of a native population were exposed for 10 generations to 10 μg/L nominal concentrations of PFOS, PFOA, and PFBS, comparable with the maximum values found in European rivers. All treatments showed reduced growth at most/several generations. No effects on survival, development, and reproduction were found. A final tolerance-induction test was performed exposing the pre-exposed experimental cohorts to 100 µg/L PFOS and 150 µg/L PFOA for a whole life cycle. Factorial analysis of variance revealed no difference between treatments (i.e., PFOS vs PFOA), indicating no induced tolerance. Instead, organisms pre-exposed to PFBS were the most stressed, followed by those pre-exposed to PFOA and PFOS, with earlier emergence and reduced adult weight. The results may be related to general stress and genetic erosion induced by long-term laboratory culture, but also to long-term toxicant exposure. However, no effects at the population level (population growth rate) were proved, and thus a toxicity risk in real ecosystems at the tested concentrations seems unlikely.
