The vasopressor activity of several orally active sympathomimetic amines was investigated in experimental studies. Compounds studied are 1-(m-hydroxyphenyl)-2-aminopropanol-hydrochloride, 1-(p-hydroxycyclohexyl)-2-amino-propane hydrobromide, N-methyl homolog of 1-(p-hydroxycyclohexyl)-2-amino-propane HBr, and the optical isomers of 1-cyclohexyl-2-methyl-aminopropane-hydrochloride. Saturation of the ring of several phenyl compounds to produce cyclohexyl isopropylamines results in compounds which possess prolonged vasopressor effects given orally. Saturation of the ring results in some diminution of vasopressor effects; the activity of the compounds ranges from 1/6 to 2/3 that of their phenyl analogs when given iv. However, these differences are less evident when the drugs are given orally. The cyclohexyl compounds tested exhibit the unexpected characteristic of being much less stimulative of the central nervous system than are their phenyl analogs.
