Abstract
The purpose of this study was to determine the mechanism by which PFDA inhibits peroxisomal β-oxidation. PFDA in corn oil was administered IP to 2 groups of female Sprague-Dawley rats at 10 and 40 mg/kg . Control rats received vehicle, while a 4th group was fed 0.01% ciprofibrate (CIP). Two weeks later, the rats were sacrificed and liver homogenates prepared. A portion of the homogenates was subjected to discontinuous sucrose gradient centrifugation for the isolation of peroxisomes. The activities of the peroxisomal β-oxidation enzymes fatty acyl CoA oxidase (FAO), enoyl CoA hydratase (HYDRA), 3-hydroxy-acyl CoA dehydrogenase (DEHYDRO), and 3-ketoacyl-CoA thiolase were significantly increased by both PFDA and CIP in both homogenates and isolated peroxisomes. Total peroxisomal β-oxidation was increased by CIP, but decreased by 40 mg/kg PFDA. Inhibition studies using control peroxisomes demonstrated that FAO was slightly inhibited by PFDA; however, the bifunctional protein activities of HYDRA and DEHYDRO were strongly inhibited, at a Ki of 5 µM. Neither CIP nor capric acid inhibited any of peroxisomal enzyme activities. These studies showed that PFDA is a noncompetitive and reversible inhibitor of the peroxisomal bifunctional protein.
