Mycelial growth inhibition of Shiitake (Lentinula edodes) by several terpenoids isolated from Sugi (Cryptomeria japonica) wood

Methanol extracts of sugi (Cryptomeria japonica D. Don) wood were separated into toluene-insoluble, hexane-toluene-insoluble, and hexane-toluene-soluble fractions. Ferruginol, sugiol, sandaracopimarinol, sandaracopimarinal, phyllocladanol, and β-sitosterol were obtained from the hexane-toluene-soluble fraction by column chromatography on silica gel. The inhibitory activity of each terpenoid on mycelial growth of shiitake (Lentinula edodes (Berk.) Pegler) was examined on a potato dextrose and agar medium at 25°C. Ferruginol, sugiol, and sandaracopimarinol were found to inhibit the mycelial growth of shiitake at a concentration of 5000 μg cm-3. The 50% inhibition concentration of ferruginol was determined at ca. 100 μg cm-3 and that of sandaracopimarinol at ca. 1000 μg cm-3. The mycelial growth inhibition of shiitake is probably due to a synergistic effect of ferruginol and sandaracopimarinol, which are the major terpenoids in sugi wood.
