Abstract
A rapid and sensitive method is described for the determination of nitrites in water. Nitrite was derivatized with 4-methyl-7-aminocoumarin in sulphuric acid medium to give the corresponding 7-diazo compound which was, in turn, hydrolysed to the highly fluorescent 4-methyl-7-hydroxycoumarin. The latter compound was detected fluorimetrically at 380 nm after excitation at 325 nm. High-performance liquid chromatography (HPLC) was required to separate 4-methyl-7-hydroxycoumarin from excess 4-methyl-7-aminocoumarin. Separation was optimized on a cyano stationary phase with apolar hexane-isopropanol (95 to 5 volume per volume) as eluent. Response curves were linear in the nitrite concentration range 1 to 30 ug per litre and the limit of detection was 0.5 ug per litre. The respective repeatability and reproducibility characteristics for 6 replicate samples of 5 ug per litre were 2.5 and 4.7 per cent.
