Abstract
IPA COPYRIGHT: ASHP A single dose study of sulpiride availability was carried out by Latin square design in 6 healthy subjects to determine the effects of concomitant administration of either sucralfate or an aluminum hydroxide-magnesium hydroxide combination. Based on 48 hr urinary excretion, a therapeutic dose of sucralfate decreased availability from diff31% to diff18.6%, while the hydroxide combination decreased availability to 20.9%. Complexation and interference with sulpiride absorption was suggested as the mechanism for the interaction. Even when sucralfate or hydroxide administration was 2 h prior to sulpiride, a 25% reduction in availability was noted. It was concluded that the interactions were clinically significant, and that sulpiride administration should precede sucralfate or hydroxide administration by at least 2 h.
