Abstract
HEEP COPYRIGHT: BIOL ABS. To produce an experimental model of cedar pollinosis and asthma, guinea pigs were sensitized with a purified cedar pollen antigen. The purified cedar pollen antigen was obtained from cedar pollen by the method of Yasueda and others. Two groups of 10 Hartley guinea pigs were immunized with i.p. injections of 1.0 mug or 10 mug of the purified cedar pollen antigen with aluminum hydroxide gel. Both of the 2 groups produced both reagninic and r1 homocytotropic antibodies. The reagin was distinguished from r1 antibody by its long persistence in homologous skin sites and its lability to heating at 56? C for 4 h. The tracheal smooth muscle from guinea pigs, which had been passively sensitized with the homocytotropic antibodies, showed contraction by the challenge of the purified cedar antigen (0.1 mug/l) in vitro. There was a significant correlation between the degree of the tracheal muscle constriction and the titers of 48 h PCA (passive cutaneous anaphylaxis) reactions (P 0.001). Guinea pigs, passively sensitized with homocytotropic antibodies, showed symptoms of pollinosis such as rhinorrhea, sneeze and epiphora, observed by the inhalation of not only purified cedar pollen extract but also cedar pollen. Bronchial constriction was induced only by inhalation of purified extract, but not by that of cedar pollens. Guinea pigs sensitized with purified cedar antigen can be a useful experimental model of cedar pollinosis and asthma.
