Abstract
The metabolism of several organic nitrates is reviewed. The isolation and characterization of organic nitrate-reductase is discussed. The metabolic pathways in the degradation of glyceryl-trinitrate (55630), isosorbide-dinitrate (87332) and pentaerythrityl-nitrate (1607007) are described. The temporal relationship between plasma concentrations and biological activity is considered. The vasodilator potency of intact organic nitrates is compared to the potency of the parent compounds. The author concludes that organic nitrates are absorbed into the portal circulation and are rapidly and completely degraded by the liver before reaching systemic circulation. Therefore, organic nitrates have little chance of producing vasodilation.
