Abstract
To clarify the critical day of teratogenesis caused by di-n-butyltin diacetate (DBTA), three-month-old pregnant rats of Wistar strain were treated orally with DBTA at different gestational stages, and sacrificed on day 20 of gestation for fetal observation. In the first experiment, organogenetic phase was subdivided into 4 periods (gestational days 7-9, 10-12, 13-15 and 16-17), and DBTA (15 mg/kg) was given to pregnant rats in each period. Treatment on days 7-9 of gestation caused external (cleft mandible, cleft lower lip, ankyloglossia and/or schistoglossia) and skeletal abnormalities (fusion of the ribs and vertebrae). In the second experiment, a single dose of DBTA (15 or 30 mg/kg) was given to pregnant rats on 3 different days (gestational day 7, 8 or 9). Treatment on day 8 of gestation caused external and skeletal abnormalities.
