IPA COPYRIGHT: ASHP Blood and urine levels of trazodone (I) and 1-(3-chlorophenyl)piperazine (II) were determined in 4 healthy humans following a single dose of 150 mg. Twenty-four h urinary excretion data showed an accumulation of 0.5% I and 0.15% II. Peak plasma levels of I were observed within 2 h, while peak levels of II amounting to 1% of I levels were observed between 2 and 4 h. Plasma half-life of II was slightly longer than I. Implications of the results regarding the role of II in the therapeutic efficacy of I are discussed.
