IPA COPYRIGHT: ASHP The pharmacokinetics of amiodarone (I) were studied in 5 cardiac patients (aged 37-67 yr) with chronic renal insufficiency undergoing regular hemodialysis. A 150 mg IV bolus of I was given, followed by a standard oral dose schedule. The regular 4 h hemodialysis regimen was started immediately before the IV bolus. Data from 5 healthy volunteers receiving the same IV dose of I and following the same sampling schedule were used for comparative evaluation. There was no apparent difference in plasma half-life between healthy volunteers and patients on dialysis, and no I was detectable in the sample of dialysis fluid or in the dialyzer membrane. It was concluded that in acute administration, dialysis treatment does not modify the kinetic behavior of I and should not therefore require any adjustment of therapy.
