Efficacy and safety of intramuscular glucosamine sulfate in osteoarthritis of the knee: randomized, placebo-controlled, double-blind study

IPA COPYRIGHT: ASHP A randomized, multicenter, double-blind study of the efficacy and safety of glucosamine sulfate was conducted in 155 patients (ages 19-75 yr) with osteoarthritis of the knee who received an intramuscular injection of 400 mg glucosamine sulfate (Dona) or placebo twice a wk for 6 wk. Responders to treatment were considered those with a reduction of at least 3 points in the Lequesne index, together with an overall judgement by the investigator. Glucosamine caused an average reduction of 3.3 points in the index, compared with 2 points for placebo. The responder rate in evaluable patients was 55% with glucosamine and 33% with placebo. Local and systemic tolerability of glucosamine was good and comparable to that of placebo. It was concluded that intramuscular glucosamine is effective and well tolerated for the treatment of knee osteoarthritis.
