Abstract
IPA COPYRIGHT: ASHP The effect of low dose aspirin (I) on platelet aggregation induced by a single or paired aggregating stimuli with arachidonic acid, platelet-activating factor and epinephrine was studied in 6 healthy men who received 20 mg of oral I shortly before midnight on 7 consecutive days. After the low dose I regimen, serum thromboxane B2 generation was reduced by 85 to 93% (from 266.8+-22.2 to 28.8+-4.3 ng/ml), and arachidonic acid-induced platelet aggregation was completely suppressed. The second wave of irreversible aggregation induced by platelet activating factor or epinephrine was also prevented. However, a full, irreversible platelet response was restored when platelet activating factor and epinephrine were used in combination. In vitro addition of 50 muM aspirin resulted in undetectable thromboxane B2 generation but had no effect on platelet aggregation induced by the combined stimuli.
