Abstract
Subjects having a history of sensitivity to toluene-diisocyanate (584849) (TDI) and with either dual or late asthmatic reactions after TDI challenge were studied to determine if treatment with ketotifen inhibits asthmatic reactions induced by TDI. Ketotifen was given to three of the patients at a dose level of 1 milligram, twice a day, for 7 days. The other three patients received treatment with a placebo. Airway responsiveness to methacholine was determined on the first and sixth days of treatment. Challenge to TDI was performed 30 minutes after ketotifen administration on day seven. Inhalation challenge with methacholine was performed at 8 hours after the exposure to TDI. The forced expiratory volume in 1 second was markedly decreased following exposure to TDI for these subjects treated either with ketotifen or with placebo. The authors conclude that ketotifen is not effective for the treatment of TDI induced asthma and they suggest it not be used in the prophylaxis of asthmatic reactions induced by TDI in sensitized subjects.
