IPA COPYRIGHT: ASHP The apparent and true partition coefficients (TPC) and pKa values of a number of amphetamine like drugs were measured. The apparent partition coefficient (APC) in chloroform-water and heptane-water was measured in Teorell buffer of various pH values such that the concentration of drug in the organic and water layer was roughly the same. Drug concentrations were measured using gas chromatography. The apparent partition coefficient at pH 7.4 and the true partition coefficient, being the partition coefficient of the neutral base, was then computed. The following equation was used: TPC = APC (1)+ 10pKa-pH It was suggested that the excretion of those drugs which are weak to moderately strong bases with a relatively high lipid solubility may be suppressed under a strong alkaline condition; this would prevent their detection in athletes using amphetamine-type drugs.
