Effects of trifluralin and oryzalin on the human erythrocyte Ca-ATPase

The effects of in-vitro exposure to the dinitroaniline herbicides trifluralin (1582098) and oryzalin (19044883) on human erythrocyte calcium-ATPase (Ca-ATPase) activity were reported. Erythrocyte membrane fragments were isolated from fresh human blood and incubated with 0.1 to 0.5 millimolar (mM) concentrations of trifluralin or oryzalin in the presence and absence of calmodulin. Magnesium-ATPase activity was inhibited 10.4%, 22.9% and 29.6% at concentrations of 300, 400 and 500 micromolar (microM) oryzalin. Trifluralin did not affect Ca-ATPase activity in the presence or absence of calmodulin. Almost 100% inhibition of Ca-ATPase activity was observed at a concentration of 0.4mM oryzalin. The concentrations required to produce a 50% reduction in Ca-ATPase activity (IC50) were calculated to be 336 and 228microM oryzalin in the presence and absence of calmodulin, respectively. The IC50s of trifluoperazine (117895) were determined to be 169 and 100microM with and without calmodulin, respectively. The different effects of trifluralin and oryzalin were discussed in terms of their molecular structure.
