Abstract
IPA COPYRIGHT: ASHP Transdermal films of ketorolac tromethamine containing both polyvinyl alcohol and povidone (polyvinylpyrrolidone) and different permeation enhancers and transdermal patches of this drug containing silicone-or acrylic-based pressure sensitive adhesives were prepared, and both the permeation of ketorolac across excised rat skin from transdermal films and the permeation of the drug across rat skin, with and without pretreatment with different volatile oils and solvents, from patches containing an acrylic-based adhesive were studied. The permeation enhancers used in the transdermal films of ketorolac included propylene glycol, oleic acid, polysorbate 60 (Tween 60), polysorbate 80 (Tween 80), and sorbitan monooleate (Span 80). The results showed that the rat skin permeation of ketorolac from films prepared without permeation enhancers was not satisfactory. The incorporation of permeation enhancers in the films enhanced the absorption of ketorolac. Also, the permeation of ketorolac across untreated rat skin from transdermal patches was not satisfactory. However, pretreatment with volatile oils dramatically increased the absorption of ketorolac.
