Abstract
A sensitive reverse phase high performance liquid chromatographic technique was developed for the quantitative determination of gossypol (303457) and other significant terpenoid aldehyde compounds in cotton dust. A solvent system comprising isopropanol, acetic-acid, water, and acetonitrile was found superior to one composed of acetonitrile, water, and acetic-acid. Neither was able to resolve hemigossypols and methylated hemigossypols from their desoxy precursors, and the latter was also unable to resolve heliocides. The concentration of gossypol in cotton dust from 2 crop years ranged between 156 and 1,323 parts per million and exhibited an inverse relationship to particle size. There was significantly less gossypol present in all the 1982 cotton dust samples compared to the 1981 specimens. The extraction efficiency for gossypol was 95 percent, and the efficiency of the entire technique in terms of the determination of a known amount of the product in the solvent was about 90 percent, yielding an estimated total efficiency for gossypol recovery of 85 percent. The values obtained in this system were higher than those reported previously using a methanol based system, where failure to maintain a high enough flow rate and a high concentration of the organic modifier methanol resulted in significant degradation of the chromatographic peaks. The authors conclude that the presently reported method is valuable for the analysis of terpenoid aldehyde compounds in cotton plants and cotton dust.
