Abstract
A simple, specific and selective HPLC method has been developed for the unified quantification of four proton pump inhibitors (rabeprazole sodium, esomeprazole magnesium trihydrate, pantoprazole sodium, and dexlansoprazole) and two gastroprokinetic agents (itopride hydrochloride and cinitapride hydrogen tartrate). Chromatographic resolution of selected analytes was achieved on a Pursuit C-18 column i.e. 4.6 mm x 25 cm, 5 mu m, maintained at 40 degrees C through isocratic elution and a detector set at preferential detection wavelength of 280 nm. The optimized mobile phase consisted of a 10 mM potassium dihydrogen phosphate buffer, acetonitrile and methanol (50:18:32, v/v). The mobile phase flow rate was adjusted at 1 mL/min and chromatographic separation was achieved in 10 min. Validation as per ICH guidelines proved the precision and accuracy of the method in the established linearity range.
