Abstract
PESTAB. A high-pressure liquid chromatography (HPLC) system was developed for complete separation of six rotenoids, using a reversed-phase gradient system. A chromatograph with a UV detector and gradient elution capacity was used. The chromatograph had a Zorbax ODS column (25 cm x 2.1 mm I.D.,), particle size 6-10 mug. The oven temperature was 34DEG and the column pressure 2000 psi. The six rotenoids tested were separated by gradient elution. The initial solvent composition was methanol-water phosphoric acid (60:39.9:0.1). The final solvent composition was methanol-water phosphoric acid (85:14.9:01.). The gradient rate was 1%/min. The order of resolution was rotenolone, tephrosin, rotenone, deguelin, dehydrorotenone, and dehydrodeguelin. Total separation of the six compounds required about 55 min. This method is practical for industrial quality control. Quantitation is possible.
