IPA COPYRIGHT: ASHP Research on the anti-inflammatory effects of prednisolone sodium phosphate (I) ophthalmic solution and prednisolone acetate (II) ophthalmic suspension is reviewed critically and methodological problems in the study of ocular drugs are identified. A pharmacokinetic model of the absorption of water-insoluble drugs (such as II) and water-soluble drugs (such as I) including drug elimination and dissolution, rates of drug penetration and transport is presented; both drugs have moderate to high anti-inflammatory effects. It is concluded that even though anti-inflammatory effects are similar, a solution of I is preferable to a suspension of II due to the possibilities of poor particle distribution leading to incorrect dosing.
