Abstract
In this study the influence of the amino- and nitroderivatives of benzene on Wistar rats and their offspring was investigated. Pregnant Wistar rats were exposed to 3,4-difluoroaniline (DFA), 3,4-difluoronitrobenzene (DFNB), 2-nitro-4-bromophenol (NBP), and 3-nitrobenzoic acid (NBA) by whole body inhalation at different concentrations (from 0 to 3 mg/m3 for 4 hr/day 7 days/week) throughout gestation. The LOAELs (Lowest Observed Adverse Effect Level) of DFA, DFNB, NBP and NBA were 0.9 mg/m3, 0.9 mg/m3, 2.8 mg/m3, and 1.0 mg/m3 for the maternal toxicity, respectively. The maternal toxic effect of the investigated compounds was characterized by changes in liver, kidneys, and central nervous system. The LOAELs of DFA, DFNB, NBP and NBA were 0.3 mg/m3, 0.3 mg/m3, 1.0 mg/m3, and 0.4 mg/m3 for the developmental toxicity, respectively. The effect of the compounds was manifested by embryonal mortality, teratogenic effects and changes in central nervous system and physical development of progeny.
