Rats. The acute LD50 of sodium tripolyphosphate (TPP) was 525 +/- 52 mg/kg. Groups of 14 male rats were maintained for 1 month on diets composed of Fox Chow meal with the following additions: group I, control--basal ration only; groups II, III and IV received 0-2, 2.0 and 10% respectively.

Slight retardation of growth was observed in rats in groups II and III and marked retardation in group IV. Kidney weights were possibly slightly elevated in group III and substantially raised in group IV. Tubular degeneration and necrosis of the kidneys were observed in rats of group IV.

Dogs. One dog fed 0.1 g TPP/kg/day for 1 month grew well and exhibited normal tissues on histological examination.

A second dog maintained for 154 days on a diet containing 1 g TPP/kg/day increasing to 2.5 g/kg/day by the end of the test, exhibited "phosphate nephritis" and left ventricular hypertrophy. Other organs and blood and urine analyses revealed no abnormality.
