Rats. The acute oral LDso of sodium hexametaphosphate (HMP) in female rats was 2900+258 mg/kg. Groups of 5 male rats were maintained for 1 month on diets composed of Fox Chow meal with the following additions: group I, controls--basal ration only; group II, controls--10% sodium chloride; group III, controls--5 % disodium orthophosphate; groups IV (a,b,c), 0.2, 2-0 and 10 % respectively of HMP.

Retardation of growth was observed in rats of groups II, IV(a) and IV(c). Normal growth was maintained in rats of groups I, III and IV(b). Growth inhibition of ra*s in group IV(a) could not be explained. Rats in group IV(c) had increased kidney weights and suffered kidney injury characterized by tubular degeneration and necrosis ("phosphate nephritis"). Some lesions of the same type were observed in group IV(b) but rtone in group IV(a).

Dogs. One dog fed 0-I g HMP/kg/day for 1 month lost an insignificant amount of weight. Urine and blood analyses and organ weights were not affected by HMP. No histological abnormalities were evident.

A second dog maintained for 154 days on a diet initially containing 1 g/kg/day, increasing to 4 g/kg/day for 1 month lost weight significantly. Increased heart weight with left ventricular hypertrophy and kidney changes ("phosphate nephritis") were the other abnormal findings.
