In an approach to combat the growing antitubercular resistance and to develop new antitubercular agents having more potent effect, here, we have synthesized some novel substituted 2,5-dimethylpyrrolyl benzohydrazide derivatives. 4-(2,5-Dimethyl-1H-pyrrol-1-yl) benzohydrazide (III), was synthesized using ethyl 4-aminobenzoate as starting material. The hydrazide III was reacted with substituted benzole acid using HBTU and DIEA in DMF to give 4-(2,5-dimethyl-1H-pyrrol-1-yl)-N'-(substituted benzoyl) benzohydrazide derivatives IV(a-g). Structures of the newly synthesized compounds were confirmed on the basis of physicochemical parameters and spectral data (infrared, H-1 NMR, C-13 NMR, and mass spectra). All the synthesized compounds were screened for their antitubercular activity using Microplate Alamar Blue (R) Assay method. Compound IVf has showed better antitubercular activity at minimum inhibitory concentration value of 12.5 mu g/ml.
