Abstract
The promutagen activating capacity of chicken S9 and mouse S9 were studied in-vitro. Leghorn-chicken livers or Aroclor-1254 induced male F1 hybrid CBA57BL-mice or uninduced mice livers were homogenized. Nicotinamide-adenine-dinucleotide-phosphate and glucose-6-phosphate were added for complete activation or omitted for nonactivation conditions. S9 mixtures were added to 2-aminofluorene (153786) (2-AF), cyclophosphamide (50180) (CP), and benzo(a)pyrene (50328) (BaP). Mutagenic activities were tested with Salmonella-typhimurium tester strains TA-1950, TA-100, and TA-98. Positive controls used were sodium-azide (26628228) or 2,7-diamino-4,9-dihydroxy-5,10-dioxo-4,5,9,10-tetrahydro-4,9-diazopyrene. A liquid suspension yeast/microsome test was performed. The recombinogenic activity of 2-AF metabolized by the S9 preparations from mouse and chicken were determined. Young male and female chicken livers were used to examine the metabolic activation of 2-AF. CP was slightly mutagenic without activation and moderately mutagenic when Aroclor-1254 induced mice or uninduced chicken S9 preparations were used. The activities of S9 preparations with BaP were similar to CP. S9 fractions from chickens were highly active in converting 2-AF to mutagenic metabolites as compared to S9 preparations from uninduced or induced mice. In the yeast test, chicken S9 was far more effective in converting 2-AF to an active species than S9 from uninduced mice. The S9 from male and female chickens possessed high activities with 2-AF. The authors conclude that S9 preparations from chicken livers activate promutagens more effectively in Salmonella-typhimurium and yeast tests than S9 from mouse liver.
