A possible connection between retained radioactivity of 2-chloroethanol (107073) in rat liver and the formation of the fatty acid conjugates 2-chloroethylstearate (1119751) (2-CES) was investigated. An authentic standard of 2-CES was prepared by heating a mixture of 2-chloroethanol and stearic-acid for 2 hours with hydrochloric-acid. Male Sprague-Dawley-rats were injected with 50mg/kg of 2-chloroethanol in mineral-oil. Five days following treatment the rats were sacrificed and microsomes were prepared from their livers. A significant drop in body weight of the rats was noted 1 and 2 days following treatment, but this weight was recovered after 5 days. The mean fresh weight of the liver was also reduced in experimental rats compared to controls. The presence of 2-CES in lipid extracts of microsomes from exposed rats was demonstrated by thin layer chromatography, high performance liquid chromatography and mass spectrometry using the standard preparation of this compound for comparison. No 2-CES was found in control rats. The authors conclude that they have demonstrated the in-vivo formation of 2-CES, a novel conjugate of 2-chloroethanol and that further studies of all possible fatty acid conjugates should be carried out.
