IPA COPYRIGHT: ASHP Production of paracetamol (acetaminophen) tablets was studied using a direct wet granulation method which employs a low melting point binding agent. Stearic acid was used as the binding agent. The binding agent melts and coats the powder particles, forming granules directly as the powder cools. Tablets produced by the method exhibited short disintegration times and rapid dissolution rates. Humidity had no effect on the properties of tablets stored for 15 months. Storage at 45DG considerably decreased the dissolution rates of all formulations. Additional excipients needed for ideal formulation, and the advantages and disadvantages of the method, are also discussed.
