This review of oncogenesis in culture traces the early stages in the history of chemical carcinogenesis, with particular emphasis on the major research of P. Yoshida and other Japanese workers in this field. The need to elucidate the cellular and molecular mechanisms whereby chemical agents such as the polycyclic aromatic hydrocarbons exert their oncogenic activity in mammalian tissues has led to the development of a number of cell culture systems which permit quantitative measurements of malignant transformation under rigidly controlled conditions. Several critical questions concerning the cellular mechanisms of malignant transformation have been asked and answered. The main part of this review deals with the work carried out in the authors' laboratory (84 references).
