Antibody Production in Human Tonsil—Detection of Ige Antibody Forming Tonsil Lymphocytes—

IgE and IgE antibody bearing lymphocytes in human tonsil were estimated by the use of isolated single cells. Most of the isolated lymphocytes were fixed with 0.1 % formalin and investigated by immunofluorescence to detect and count total immunoglobulins, IgE and total mite (Dermatophagoides farinae) antibodybearing lymphocytes. Sheep red blood cells (SRBC) coupled with anti-IgE was reacted with fixed tonsil lymphocytes. SRBC-anti IgE bound lymphocytes were obtained by refractionation with 9 % Ficol-Urographine and stained by directly FITC-mite conjugate to detect IgE antibody cells. Effects of formalin fixation to lymphocyte were as follows; 1) SRBC-RFC was not formed at all after the fixation. 2) Population of immunoglobulin-bearing lymphocytes was not significantly influenced. 3) Serum IgE antibody activity was not significantly changed. Percentage of IgE lymphocytes to total cells were 0.21–0.78, and that of anti-mite IgE antibody cells to IgE lymphocytes was 1.4–6.4. From these results, it is concluded that IgE population determined by the immunofluorescent staining of single cells was very high, and antimite IgE antibody-bearing lymphocytes were also demonstrated.
