Abstract
Burns and Sandford recently reported some interesting data about the sensitivity of human herpesvirus 6 (HHV-6) to antiviral drugs in vitro. Their findings concur with results we obtained using slightly different experimental procedures. Briefly, in our experiments, peripheral blood lymphocytes (PMNL) from a healthy donor were infected with HHV-6 strain SIE at a multiplicity of 0.02 TCID sub(50)/cell and incubated in the presence of various concentrations of acyclovir, ganciclovir, and phosphonoformic acid (PFA; foscarnet). After infection for 6 days, the multiplication of HHV-6 was quantified by means of cytopathic effect scoring, DNA hybridization in culture supernatant pellet, and indirect immunofluorescence assay (IFA) on fixed cells using a reference HHV-6-positive human serum. The three quantification techniques gave concordant results and allowed us to determine the 50% and 90% inhibitory concentrations (IC sub(50) and IC sub(90)) for each antiviral tested. When using IFA, the IC sub(50)s were 27 mu M, 1.1 mu M, and 2.7 mu g/ml for acyclovir, ganciclovir, and PFA, respectively.
