Abstract
The effects of several steroids and gonadotropins on the maturation of follicle-enclosed oocytes of the catfish, Clarias batrachus were examined in vitro. All the steroids tested (pregnenolone, 17-alpha-hydroxy-pregnenolone, progesterone, 17-alpha-20-beta-dihydroxy-4-pregnen-3-one, cortisol, corticosterone, 11-deoxy-corticosterone, 11-deoxycortisol, androstenedione, 11-keto-testosterone and testosterone) except the estrogens were found to initiate final oocyte maturation as evidenced by germinal vesicle breakdown (GVBD). 17-alpha-20-beta-dihydroxy-4-pregnen-3-one (17-alpha-20-beta-DHP) appeared to be the most effective in inducing GVBD among all the steroids tested. Progestins were more effective than corticosteroids. Androgens were effective in inducing GVBD at their higher concentrations only (1.0, 10 and 100-mu-g/ml). Whereas, the corticosteroids and progestrogens could induce GVBD at a ten times lower dose level than that of androgens (0.1-mu-g/ml). Amongst the androgens tested, testosterone was the most effective followed by androstenedione and 11-ketotestosterone and in the corticoids, 11-DOCA was most potent and cortisol was least effective in eliciting GVBD response. Amongst the gonadotropins tested, Mystus gonadotropin (mGtH) was most effective followed by Ovine Luteinizing Hormone (oLH) and Ovine Follicle Stimulating Hormone (oFSH). Although, at the lower dose levels (1.0, 10-mu-g/ml) no appreciable difference was noticed between oLH and oFSH response, but at 100-mu-g/ml dose level oLH was more effective than oFSH.
