The teratogenic effects of theophylline in mice

Pregnant albino mice were administered single i.p. injections of the methylated xanthine theophylline at dosages of 100,150 or 200 mg/kg on days 10 through 13 of gestation. Under these conditions, theophylline was moderately embryolethal but highly teratogenic with cleft palate, ectrodactyly, syndactyly and micromelia as the predominant malformations. The peak embryotoxic response occurred in fetuses surviving treatment on day 11. A left-sided preponderance of limb defects was also observed.
