A reversed phase high performance liquid chromatographic( RP-HPLC) method with polystyrene-divinylbenzene (PS-DVB) resin(MKF-RP-MH) was developed for the determination of thymosin alpha(1). In the method, thymosin alpha(1) was chromatographically analyzed with a mobile phase A:0.01 mol/L phosphate buffer solution( pH = 7.0 or 8.0) and B: acetonitrile, linear gradient elution condition: 0-20 min, 0%-10% acetonitrile, 20-40 min, 10%-30% acetonitrile; column temperature was set at 30 degrees C; flow rate was 0.8 mL/min; The detection wavelength was 214 nm. The standard sample and two kinds of thymosin a, samples were analysised under the optimum chromatographic condition. The calibration curve was in good linearity within the range of 0.05-4.0 g/L with the regression equation of C = 0.0099A-0.0798 (r = 0.9994). The maximum amount of thymosin alpha(1) on MKF-RP-MH column was 120 mu g (6 g/L).
