Abstract
Paper chromatographic studies were made on (a) plasma from rabbits given injections of methyl- or carbonyl-C14-labeled urethan 2½ and 3½ hours previously, (b) mouse plasma obtained 2½, 6, or 12 hours after injection of urethan-C14, and (c) urethan-C14 solutions in which mouse lung, liver, and skin had been incubated. Precautions were taken against loss of urethan or of similarly volatile compounds. The methods developed included a procedure for the direct hydrolysis of carbonyl-C14-urethan to C14O2 without elution from the paper strip. In all cases only a single migrating component was found, whose rate of migration and volatility corresponded to those of free urethan. This study, therefore, suggests that there is no specific metabolite of urethan which could be involved in urethan carcinogenesis. © 1960, American Association for Cancer Research.
