Abstract
Adsorption HPLC (high-performance liquid chromatography) with a slurry-packed calcium hydroxide (lime) column was used to separate cis/trans isomers of alpha- and beta-carotene. The mobile phase consisted of hexane and acetone in the proportions 99:1 or 99.5:0.5 (v, v). Isomers were prepared from standards by photoisomerisation catalysed with iodine. Identification of the isomers was achieved by measuring the UV/visible absorption spectra obtained with a diode array detector (DAD) during HPLC. Signals could be recorded at seven different wavelengths simultaneously. Chromatographic signals were recorded at 328 nm and 442 nm for alpha-carotene and 335 nm and 450 nm for beta-carotene. Applications to foods were carried out. Carrot juice was extracted with acetone:ethanol without saponification and several carotenoids as well as cis/trans isomers of alpha- and beta-carotene were detected. The extraction was optimised for use on vegetables and other food with low fat content.
