1. 1. A method for the application of FDNB as a cytochemical reagent is described. The tissue sections are reacted with FDNB in aqueous ethanol containing sodium bicarbonate and the nitro groups of the treated sections converted into a suitably coloured derivative by reduction, diazotisation and coupling with a phenol. 2. 2. From a survey of a wide range of tissue sections it was observed that when used alone, FDNB produces a dense overall reaction, but when it is preceded by the blocking agent benzoyl chloride, the reaction is confined to cell nuclei and a few characteristic cytoplasmic sites. The conditions under which this reaction after blocking takes place and the mechanism of the reaction are discussed. © 1961.
