The seed oils of Paeonia ludlowii in Tibet were extracted by ultrasound-assisted N-hexane extraction method, Orthogonal tests were carried out based on individual factor experiment, the main physicochemical indexes of Paeonia ludlowiis seed oil were determined, at the same time, the fatty acid compositions were analyzed by GC-MS. The results indicated that the yields of Paeonia ludlowiis seed oil were (39.51±1.04)% and (28.26±1.22)%, The optimal oil extraction process were obtained as follows: extraction temperature 60 â, ultrasonic time 50 min, ultrasound power 160 W, sample size 100 meshes, liquid-solid ratio 12 mL/g. The physicochemical indexes of Paeonia ludlowiis seed oil were: iodine values (153.234±2.963) g I/100 g, acid values (1.521±0.062) mg KOH/g, saponification values(186.398±3.531) mg KOH/g, peroxide values (1.406±0.069) mmol/kg, refractive index(1.470±0.013) nD 20 â, which were accordance with the grain industry standard of LS/T 3242-2014.The content of γ-tocopherol was 550.27 mg/kg in seed oil, wherein, β-sitosterol was 4.94 μg/mL, Fucosterol was 4.33 μg/mL and squalene was 153.67 μg/mL.Total 20 fatty acids were detected in Paeonia ludlowiis seed oil, the main fatty acids were oleic acid, linolenic acid, linoleic acid and palmitic acid.There were 6 unsaturated fatty acids and the contents were 87.44%, in which there were 3 monounsaturated fat acids accounting for 41.40%, 3 polyunsaturated fatty acids accounting for 46.04%, wherein, linolenic acid accounting for 29.90% and saturated fatty acids accounting for 12.56%. © 2020, Dept. of JCCOA.
