Abstract
In view of cutaneous adverse effects of ion exchanger-treated culture medium (ACCM-II) and mycelial extract (ACME-II) of Albatrellus confluens, their primary and cumulative irritations, sensitization, phototoxicity and photosensitization in guinea-pigs and their eye mucosa irritation in rabbits were investigated. Six animals per group were used for each experiment. 1) The skin primary irritation test was carried out by their 24 hours' application with closed patch on lateral abdomen. No skin reaction to ACCM-II and ACME-II was observed from 24 to 96 hours after their treatments. 2) The skin cumulative irritation test was carried out by their consecutively daily application on the constant skin area during 21 days. Skin reaction was observed neither in ACCM-II nor in ACME-II groups. In histological findings, slight thickening of epidermis was observed at the applied sites of ACCM-II and ACME-II as well as solvent, and any other changes were observed neither in epidermis nor in dermis. 3) The skin sensitization test was carried out by maximization test. Erythema was observed in all the 6 positive control animals treated with dinitrochlorobenzene. Skin reaction was observed neither in ACCM-II nor in ACME-II groups from 24 to 96 hours after their treatments. 4) The phototoxicity test was carried out by the method of Morikawa et al. Erythema was observed in all the 6 positive control animals treated with 8-methoxypsoralen. No skin reaction was observed in both ACCM-II and ACME-II groups from 24 to 96 hours after their treatments. 5) The photosensitization test was carried out by adjuvant strip method. Erythema was observed in all the 6 positive control animals treated with 6-methylcoumarine. No skin reaction was observed in both ACCM-II and ACME-II groups from 24 to 96 hours after their treatments. 6) The eye mucosa irritation test was carried out by Draize's method. On the cornea, iris and conjunctiva, no irritation was observed in both ACCM-II and ACME-II groups from 24 to 72 hours after their treatments. The above results without skin irritation, skin sensitization, phototoxicity, photosensitization and eye mucosa irritation of ACCM-II and ACME-II suggest the safety for their cutaneous applications.
