Abstract
1. 1. A liver fraction which catalyzes the oxidation of 3-hydroxyanthranilic acid to 2-amino-3H-phenoxazin-3-one-1,9-dicarboxylic acid has been isolated from poikilothermic vertebrates. 2. 2. With the use of standard protein fractionation procedures, a 45-fold purified preparation was obtained. 3. 3. The fraction's activity was inhibited by Fe2+, CN-, N3-, ethylenediaminetetraacetate and α,αâ²-dipyridyl (1 mM); however, heavy metals such as Mg2+, Cu2+, Mn2+, Mo2+ had no effect on activity. The pH optimum was around pH 7.4. 4. 4. Six o-aminophenols were studied as possible substrates; with the exception of 2-amino-3-hydroxytoluene all were converted to phenoxazinones. 5. 5. Stoichiometric studies have established that 3 atoms of oxygen are consumed in the formation of 1 mole of phenoxazinone from 2 moles of o-aminophenol. © 1965.
