Abstract
Callus was initiated from crozier explants of Diplazium cognatum cultured on haft strength MS agar medium supplemented with 3 % sucrose and varied concentrations of 2,4,5-trichlorophenoxyacetic acid. Maximum semifriable callus developed and pieces of calli subcultured for 6 weeks in a medium containing 4.9 μM 2-isopentenyladenine showed 70% shoot formation. Shoots (7 cm long) were separated, rooted in haft strength MS basal agar medium in 12 weeks, hardened for 4 weeks and established in polycups containing 1:1 sand and garden soil. The micropropagated plants were free from morphological and growth abnormalities.
