OBJECTIVE: Recurrence and metastasis are the major factors affecting the curative effect of lung cancer. Research shows that heat shock protein 90α(HSP90α) is highly expressed in lung cancer cells. This study aimed to explore the effects of tobacco carcinogen BaP(Benzopyrene) on the proliferation, migration, and invasion of silencing HSP90α human lung adenocarcinoma A549 cells. METHODS: A549 cells line was cultured and divided into two groups without transfection (Control group without BaP treatment and BaP group with BaP treatment) and four groups with transfection (NC group and HSP90α-shRNA group without BaP treatment, and NC+BaP group HSP90α-shRNA+BaP group with BaP treatment). A549 cells transfected by RNA interference technique. HSP90α mRNA expression was detected by q-PCR; CCK-8 detected the effect of BaP on the proliferation of A549 cells. Transwell detected the effect of BaP on the migration and invasion of A549 cells. RESULTS: One recombinant plasmid that interfered with HSP90α expression was successfully constructed. After transfecting shRNA plasmid for 24 h, HSP90α mRNA expression level was 0.29±0.07, which was significantly lower than that of the Control group(F=38.17, P<0.05); The proliferation ability of the NC group(0.98±0.04) and HSP90α-shRNA group(0.97±0.07) were no statistically significant differences compared to the Control group, F=0.24, P>0.05. Compared with the Control group, the proliferation capacity of the BaP group(1.35±0.21) was statistically significant differences, t=2.88, P<0.05. The NC+BaP group(1.28±0.13) was compared with the NC group (0.98±0.04), the proliferation capacity was statistically significant differences, t=3.84, P<0.05. Compared with the HSP90α-shRNA group(0.97±0.07), the proliferation capacity of the HSP90α-shRNA+BaP group(0.90±0.07) was not statistically significant differences(t=0.94, P>0.05); Compared with the Control group (113±20), the migration capacity of the BaP group (180±21) and the HSP90α-shRNA group(33±8) was statistically significant differences, with the t values were 3.83 and 6.14, respectively, P<0.05. Compared with the HSP90α-shRNA group (33±8), the migration capacity of the HSP90α-shRNA+BaP group (69±12) was no statistically significant differences (t=2.56, P>0.05); Compared with the Control group (213±23), the difference in cell invasion capacity of the BaP group (738±35) and the HSP90α-shRNA group (26±5) was statistically significant, with t value were 21.11 and 13.63, respectively, P<0.05, while that of the HSP90α-shRNA+BaP group (53±10) compared with the HSP90α-shRNA group (26±5), there was no statistically significant differences in cell invasion capacity (t=2.42, P>0.05).
