RP-HPLC Determination and Pharmacokinetic Studies of Rutin and Quercetin in Rat Plasma Dosed with Sendeng-4 Decoction

A RP-HPLC method for simultaneous determination and pharmacokinetic studies of rutin and querceitn in rat plasma was developed. Plasma samples of rats were pretreated by protein precipitation with methanol-acetic acid (9:1, v/v). The analysis was performed on a hypersil BDS C(18) analytical column (250x4.6mm i.d., 5 mu m particles) with mobile phase consisting of methanol-water-phosphoric acid,47:53:0.02 (v/v). The UV detector was set at 360nm. Vanillin was used as the internal standard (IS). The linear ranges of the calibration curves were in the range 0.068-8.60 mu g mL(-1) (r(2)=0.997) for rutin and 0.052-6.5 mu g mL(-1) (r(2)=0.996) for quercetin. The mean extraction recoveries of rutin at three concentrations were 89.7, 87.6, 85.2%, respectively and of quercetin were 87.1, 84.3, 88.1%, respectively. The intra- and inter-day precisions (R.S.D) were <= 7.4% for rutin and <= 6.7% for quercetin, respectively. The lower limit of quantification was 0.068 for rutin and 0.052 mu g mL(-1) for quercetin. The validated method was used to pharmacokinetic study of rutin and quercetin in rat plasma after oral administration of Sen-deng-4 decoction. The plasma concentrations of rats showed a C(max) of 15.49 +/- 0.861 mu g mL(-1) at T(max) of 7.0 +/- 0.75 h for rutin and a C(max) of 10.65 +/- 0.658 mu g mL(-1) at T(max) of 7.3 +/- 0.63 h for quereetin.
