The in-vitro metabolism of phthalate-esters (PAE) by liver and kidney homogenates from rats, biliary excretion in Wistar-rats after administration of carbon-14 labelled di-n-butyl-phthalate (84742) (DBP), and identification of its metabolites in bile were studied. When incubated with rat liver homogenates at 37 degrees-C, added PAE was degraded at varying rates. Studies with diethylhexyl-phthalate (117817), di-n-octyl-phthalate (117840), dimethyl-phthalate (131113) (DMP), and DBP revealed that PAE having lower molecular weights were metabolized at a faster rate in rat liver, and the difference in the metabolic rates was due to the difference in their chemical structures. The rate of degradation of PAE by rat kidney homogenates was slower than in rat liver homogenates. Again, DMP, with the lowest molecular weight, was degraded at the fastest rate. The excretion of radioactivity in the bile after intravenous injection of DBP was relatively rapid, and somewhat slower after oral administration. Thin layer chromatography of the ether extract from rat bile identified four or five distinctive spots. Three of them were identified as DBP, monobutyl-phthalate (131704) (MBP) and phthalic-acid (88993). Identities of the compounds were confirmed by gas liquid chromatography.
