Abstract
Helicobacter Pylori(HP) is closely associated with duodenal ulcer in
humans. It has strong urease activity and produces ammonia on the
surface of gastric mucosa. In the previous studies we demonstrated that
HP infection in man resulted in a significant decrease of somatostatin
content in antral mucosa (Gastroenterology 1992,102(4),A80). The
purpose of this study is to examine how the G-cells and D-cells were
affected by the chronic exposure to ammonia induced by HP infection.
"'""'"""'""""'' 0.015% and 0.10% NH3 was administered to male Wistar rats
weeks. 0.3mM NaOH which had the same pH of O.Ql5% NH3 was
used as control to exclude the influence of alkalinity. We examined
l)serum gastrin levels, :Z)antral gastrin and somatostatin content,
3)population of antral G- and D-cells, and 4)antral mRNA levels of gastrin
and somatostatin. Serum gastrin levels, antral gastrin and somatostatin
content were measured by radioimmunoassay. The number of G- and Dcells
in antrum were counted in ten glands per field and the mean was
calculated from five fields. Antral mRNA levels were determined by
Nothern blot analysis and densitometry scanning.

No statistical significance was found in serum gastrin levels. 0.3mM
NaOH had no influence on G- and D-cells. mRNA levels of gastrin were
increased and those of somatostatin showed no significant change in
0.015% NH3 group compared with 0.3mM NaOH group. Conclusions:
These data indicated that the decrease of antral somatostatin was mainly
due to the decrease of population of D-cells induced by ammonia
administration, The low somatostatin levels in human antrum caused by
HP infection is largely due to the effect of ammonia it produces.
