Abstract
FRG-8813 is a histamine H,-antagonist with potent antisecretory and gastroprotective effects. The aim of this study was to examine the property of its protective effect against ammonia-induced mucosal lesion. Methods; (1)Male S.D. rats (180-200g) fasted overnight were
treated orally with FRG-8813 30 min before the administration of
1% ammonia. Gastric lesion (mm') was counted 1hr later. In time course study, rats were killed 5, 30 and 60 min after the ammonia treatment, respectively. (2)In order to measure transmucosal potential difference (PD), the stomach of anesthetized rat was mounted on a Lucite chamber. FRG-8813 (0. 1-3 mg/kg) was applied into the chamber for 30 min. The stomach was exposed for another 10 min to 0.25% ammonia after the removal of FRG-8813. After the removal of ammonia, the
perfusion systew was resumed.
Results; (1)FRG-8813 produced a dose dependent gastroprotective action. The EDs 0 value (60 min) was 3.3 mg/kg p.o. The data of the time course study were as follows; the protective effect was not observed 5 min later.

(2)The application of FRG-8813 failed to prevent the abrupt PD reduction by 0.25% ammonia at any doses, but it significantly promoted the recovery of the PD reduction dose-dependently. Conclusion: These results suggest that FRG-8813 showed potent gastroprotective effect of the ammonia lesion and the protective effect of FRG-8813 may be attributable to the acceleration of mucosal regeneration after damage.
