When Spirogyra varians was cultured at low temperature (4C) the antioxidant activity was increased over time course. The antioxidant was extracted using methanol. HPLC analysis of the extract showed four new compounds produced during cold acclimation process. The radical scavenging activities of the isolated compounds were as high as ascorbic acid. Purification of the compounds was performed and the possible structures were predicted using H-NMR, C-NMR and Mass spectrometry. The compounds were derivates of galloyl glucose and gallic acid, which is produced through shikimate pathway. A shikimate pathway related gene involved in the cold acclimation process of Spirogyra varians was also isolated and its possible role for the production of the antioxidant compounds was analyzed. The galloyl glucose derivates could be used as anti-microbial and anti-cancer agents.
