Abstract
A method was presented for determining the extent of contamination of blood by persistent organochlorine pesticide residues and for determining relative amounts in adipose tissue and biological fluids. Samples of bovine whole blood or blood serum incubated with a standard pesticide mixture were extracted with acetic-acid, then extracted with petroleum-ether and acetone. Concentrated extract was cleaned up with concentrated sulfuric-acid and further extracted prior to analysis by gas liquid chromatography with electron capture detection. For 13 pesticides, recoveries were around 90%. Standard deviations and coefficients of variation showed excellent repeatability. Similar values were obtained for whole blood and serum. Acetic-acid extraction provided hydrolization of residues from complexes with plasma proteins, while sulfuric-acid clean up precipitated fats and removed interfering compounds such as phthalate esters. The authors conclude that this method is rapid, low cost and semimicro and allows for determination of bound and free residues, providing a truer measure of human body fluid contamination by persistent organochlorine pesticides.
